By contrast, under high shear conditions it exhibits a low viscosity. The viscosity changes immediately when the shearing conditions change, for example, when the solution comes to rest after being pumped.

Welan gum microbial production characterization and applications

Microbial exopolysaccharides are of high molecular weight, environment friendly valuable natural polymers, having applications in diverse areas such as food industry, pharmaceutical industry, cement systems and cosmetics industries. Welan gum, a microbial product holds an important place among the exopolysaccharides due to its novel properties and potential applications.
The biopolymer welan gum is synthesized by the fermentation process mainly by the Alcaligenes sp., and is composed of polymer of tetrasaccharide backbone chain containing l-mannose, l-rhamnose, d-glucose, and d-glucuronic acid. It acts as a thickening, suspending, binding, emulsifying, stabilizing and viscosifying agent. It has important commercial applications in the cement systems. This review is primarily focused on the microbial production, purification, recovery, and the characterization based on the available published literature on the welan gum. Biosurfactants may be involved in pathogenesis

Bio surfactants may be involved in pathogenesis due to their surface activity; however, for security and regulatory reasons, production strains should be nonpathogenic. In the above mentioned example of rhamnolipids this is not the case as Pseudomonas aeruginosa, the most common producing bacteria, is a pathogen.

A variety of methods for the screening of biosurfactant producing microbes has been developed and successfully applied. Since the 1970th there have been various trials in this field. These screenings have mostly been limited to a manageable number of samples. In recent years automation and miniaturization have led to the development of high throughput methods for screening of biosurfactant producing strains. A broad application of such methods could eventually lead to the desired upsurge of new commercially interesting strains.
Biosurfactants may be involved in pathogenesis

An efficient screening strategy is the key to success in isolating new and interesting microbes or their variants, because a large number of strains needs to be characterized. A complete strategy for screening of new biosurfactants or production strains consists of three steps: sampling, isolation of strains and investigation of strains. Theses steps will be addressed in the next paragraphs. Bioinformatical approaches like homology search are not included herein.

In this study, the bacterium Bacillus licheniformis has been isolated from oil reservoir; the ability of this bacterium to produce a biosurfactant was detected. Surface properties of the produced biosurfactant were confirmed by determining the emulsification power as well as surface and interfacial tension. The potential application of this bacterial species in microbial-enhanced oil recovery (MEOR) was investigated. The percent of oil recovery was 16.6% upon application in a sand pack column designed to stimulate an oil recovery. It also showed antimicrobial activity against the growth of different strains of SRB (sulfate reducing bacteria). Results revealed that a complete inhibition of SRB growth using 1.0% crude biosurfactant is achieved after 3 h.

The flow behavior and dynamic viscoelasticity of welan gum solutions were measured with a rheogoniometer. The welan gum showed shear-thinning behavior at a concentration of 0.1%, but plastic behavior above 0.3% at 25°C. The dynamic viscoelasticity increased with increasing concentration, and was scarcely changeable with increasing temperature even at 80°C. Gelation did not occur even in a polysaccharide concentration of 1.0% at low temperature (0°C). An increase of the dynamic modulus was not observed on the addition of CaCl2 (6.8 mm). The dynamic viscoelasticity of welan gum solution was scarcely changeable in a wide range of pH from 2 to 12. The dynamic modulus was also scarcely changeable on addition of urea (4.0 m). Possible mode of intramolecular associations between the OH-4 of the d-glucosyl residue and the adjacent hemiacetal oxygen atom of the l-rhamnosyl residue, and between the methyl group of the l-rhamnosyl residue and the adjacent hemiacetal oxygen atom of the d-glucosyl residue were proposed.

This review includes environmental applications of these biosurfactants for soil and water treatment. Biosurfactant applications in the environmental industries are promising due to their biodegradability, low toxicity and effectiveness in enhancing biodegradation and solubilization of low solubility compounds. However, more information is needed to be able to predict and model their behaviour. Full scale tests will be required. The role of biosurfactants in natural attenuation processes has not been determined. Very little information is available concerning the influence of soil components on the remediation process with biosurfactants. As most of the research until now has been performed with rhamnolipids, other biosurfactants need to be investigated as they may have more promising properties.
